Many chemical transfection reagents have an ideal time window in which an optimal-diameter transfection complex is created. Depending on the quality of the reagent, this is usually between 5 and 30 minutes. In general, transfection reagents must be in contact with cells for a period of time before introducing some additional medium or removing the medium (to help minimize toxic effects of the reagent). The optimum time for transfection depends on the cell line, the transfection reagent and the nucleic acid used.
